We have performed germline MLH1 hypermethylation testing for Lynch syndrome in >400 individuals. Using long read sequencing, we have identified MLH1 promoter mutations associated with Constitutional MLH1 promoter hypermethylation in one of the largest studies to date!

2 patients. Same surgery. One goes home same day. The other needs APS, admission, multimodal analgesia beyond opioids, and slower return to work. Why? One answer may be RUNX1 genetics. Our collab @HotchkissBrain @UCalgary with Dr. Tuan Trang and @jeffbiernaskie shows that RUNX1, via microglial regulation, shapes opioid analgesic response, withdrawal and links biology to real world perioperative pain variability. Big step towards predicting opioid responses and developing personalized pain plans!
