Very excited to share this paper that our @valarlabs team published: We extended our prior work with the Computational Histologic Artificial Intelligence (CHAI) platform, which analyzes solely histomorphic features ("histomics"), to develop and validate a biomarker that predicts optimal first-line chemotherapy for patients with advanced pancreatic cancer, often considered to be the most technically and clinically challenging cancer. Histomics is an often overlooked yet powerful modality that can be leveraged for precision medicine. More exciting work to come!

Thrilled to share my PhD paper, now published in @NatureGenet! We used single-nucleus DNA sequencing to unravel the complex genomic evolution of pancreatic cancer, as illustrated by the cover art by my amazing mentor @ciacobu. #Genomics #PancreaticCancer nature.com/articles/s4158… Pancreatic cancer is notoriously difficult to treat. Several precision medicine opportunities are on the horizon. but we still do not fully understand the cancer's molecular vulnerabilities. Bulk sequencing gives a crude picture, but we need to see the cancer at single-cell level to further understand and treat it. #PrecisionOncology #CancerEvolution In our study, we analyzed over 137,000 single nuclei from 24 patients with pancreatic cancers in various clinical contexts, and mapped out their evolutionary trajectories at high-resolution. Key finding 1: In tumors with KRAS mutations (the most common driver in pancreatic cancer ), we saw a varied dependence on this gene late in tumor progression. This could explain why KRAS inhibitors work for some patients but not others. #KRAS #TargetedTherapy Key finding 2: For patients with inherited BRCA2 mutations, we discovered diverse ways the cancer gets rid of the remaining healthy copy of the gene. This shapes the tumor’s evolution and can impact therapy response. #BRCA #HereditaryCancer Key finding 3: We found that the inactivation of the TGF-β pathway, which normally suppresses tumor growth, occurs late in tumor development and is constantly selected for as the tumor interacts with associated microenvironment during invasion and metastasis. #Metastasis Our work was enabled by several technical advances including single-nucleus DNA sequencing and accompanying computational analysis tools. Incredible dataset mapping evolutionary trajectories in this cancer.
