Is that difference real, given that most of that difference is due to the decline in the placebo group? Especially as many other measures of muscle performance did not improve? And yes Charles, I have patients with PAD. The clinic I work in is a semi-rural, where most patients have a poor socioeconomic status. Diabetes and IHD are rampant, as is PAD. Pushing NR supplements on this patient demographic, who can barely make financial ends meet, based on unconvincing results, is highly unethical

I appreciate your points, but this study is weakly positive at best. NR showed a mere 2% improvement in baseline 6-min walk, while other muscle performance measures remained unchanged. The clinical relevance of such small gains is highly questionable. Let's not overstate the findings. Your comment, "& it showed that resveratrol degraded beneficial effects of NR" is reaching. The differences in the walk test and the groups are small.

@BradStanfieldMD Dr Stanfield I’ve seen you be pretty even handed in the past. But downplaying a 2% gain vs a 3% loss (that’s a total delta 5%) walk distance in just 6 months, doesn’t seem like something to totally disregard. Also seems like you’re overstating the hype here (what hype?).

@bryan_johnson I think you once said you can run successful N=1 trials, to me this supports that and is pretty neat. You had reasons why you tried HGH, and now you’re course correcting based on evidence. To me this says that you’re just willing to risk trying stuff for the benefit of others.

Discontinued HGH therapy 5/31 after 110 days bc side effects: symptoms suggestive raised intracranial pressure as exp previously, 15% HRV drop, headaches, increased blood glucose. Completed before & 100 day thymus MRI, results being analyzed. Evaluating other thymus rejuv option

@RaisingNad yes we got human data showing very substantial improvement in skin elasticity as well as scoring of skin appearance (fine lines) topical NR in the NR lab cream and ampoule work really well more on these products soon

The vast majority were driving a sensible speed, although some were following like it was dry pavement. Then a very small minority were passing and going way too fast. So much that I was not surprised at all when this happened.

@bryan_johnson But probability of succeeding in your main 4 areas isn’t linearly related to motivation. Often times a little nudge can push you into the success zone. I take issue with the term “magic pills” but could supplements be a nudge factor that could make the difference for many people?

@mkaeberlein To recap: you think all the good data on NR isn’t enough to justify taking it. But you also think it is enough to justify very long and large scale (aka very expensive) trials? Did people start taking 1g of vitamin C per day before multi-year trials published at that dose?

Maybe a good trial on NR or NMN will be published in the next month, year, or decade showing impressive results. If so, I'll adjust my position. Long term safety will take awhile. Probably very low risk, but you are likely perturbing a central metabolic cofactor, so who knows.

Don't think so. Just facts. Nicotinamide riboside heavily marketed to consumers for a decade without any well-powered, medium- or long-term clinical trials to support claims being made. Less data on NMN. Assumed safe (GRAS) but not really known. Most people don't know that.

The FDA has just released a statement banning NMN supplement sales. It seems as though a company called Metro International Biotech (co-founded by Dr David Sinclair) lobbied for NMN to be banned, so they can sell their proprietary blend of NMN called MIB-626
