Interestingly, immunogenic LNPs retain delivery efficacy upon repeat dosing because they suppress anti-PEG IgM levels by prompting cytokine release.

We report efficacious structures for delivery to placental trophoblasts, endothelial cells, CD4 T cells, and NK cells. Additionally, we show that inflammatory LNPs hinder neonatal growth. We believe this finding will be critical for designing pregnancy-safe RNA therapies.
