Thrilled to share our latest work identifying that #SARSCoV2 #Omicron variants harboring an L260F mutation in NSP6 have enhanced replication and pathogenesis. A not so brief thread 🧵

Today we report single-cell APEX-seq (scAPEX-seq) — a new method for unbiased mapping of *subcellular* transcriptomes at single-cell resolution. This approach reveals cell states that are not detectable by standard scRNA-seq, and enabled us to identify regulators of CAR T function that improve solid tumor killing. biorxiv.org/cgi/content/sh…

The first time you start a research lab, there’s no manual. So I made one. A practical toolkit for postdocs and early-career faculty launching their first lab: hiring, startup planning, collaborations, and building a sustainable research program. zenodo.org/records/188835…

Thrilled to share the first major preprint from the lab. Viruses are classically viewed as targets of host sensing. Do viruses also sense and respond to the host? We propose viruses may act as “biosensors” of the host signaling state. A thread👇🏾 biorxiv.org/content/10.110…

SARS-CoV-2 continue to surprise us with evolutionary jumps. In this pre-print, we describe the Identification and genomic characterisation of BA.3.2: a highly divergent BA.3-related SARS-CoV-2 lineage from southern Africa. A thread 🧵 medrxiv.org/content/10.648…

Excited to share our new work on immune aging! We explored whether the liver could serve as a temporary "factory" to produce immune factors that decline with aging, potentially helping to rejuvenate aged immunity. @mircoscopy.

With 30 new BA.3.2 sequences from the Netherlands and Denmark uploaded today, this is beginning to have a look of inevitability about it. Still waiting for a recombinant that restores ORF7a, ORF7b, and ORF8 (all entirely deleted in BA.3.2).
Marc Johnson@SolidEvidence

Three more BA.3.2 detections from US wastewater. Rhode Island (different sewershed, but near the previous one), Vermont, and Florida. All from CDC NWSS data. Looks like it might be getting a foothold in New England. @LongDesertTrain @siamosolocani @snpoehlm

Today in @Nature, in work led by @aditimerch, we report the ability to prompt Evo to generate functional de novo genes. You shall know a gene by the company it keeps! 1/n

Today in @Nature we report a new prime editing strategy that can rescue a common cause of many genetic diseases in a disease-agnostic manner. This approach converts a redundant endogenous tRNA into an optimized suppressor tRNA, enabling a single prime edit to rescue premature stop codons across different diseases. So awesome to have this great paper from @SamReffsin and Sara Cherry out! In it, we use retrospective clone tracing to show that there are particular single cell states that are more susceptible to viral infection (both SARS-CoV-2 and flu)! cell.com/cell/fulltext/…

Excited to share @Nature: How does naloxone (Narcan) stop an opioid overdose? We determined the first GDP-bound μ-opioid receptor–G protein (wt) structures and found naloxone traps a novel "latent” state, preventing GDP release and G protein activation. 🧵 nature.com/articles/s4158…
