A new paper from @IvanAhelLab about enzymes that efficiently ubiquitylate DNA and RNA in vitro and maybe in cells. With contribution from Orléans by @vvinz and a little bit myself. Part of a larger project on noncanonical Ub by DELTEX E3 ligases led by Kang Zhu as the 1st author.

@Cochrane_lab Also, be aware that phosphonium-based coupling agents (like PyAOP) react with phenols. Not aminium-based ones (like HATU).

@Cochrane_lab Free phenols are usually not a problem (but I never worked with dopamine), but phenolic esters are stable to TFA cleavage. In the case of 2-hydroxy-5-nitrobenzyl and 2-hydroxy-4methoxybenzyl (Hmb backbone amide protecting group), a piperidine treatment cleaves the esters.

Peptide synthesis question. To couple dopamine to a peptide acid, do OH groups on dopamine need to be protected? Presumably as long as we activate acid first, then chuck in dopamine, amine reacts fastest and any unwanted phenolic ester gets hydrolysed in TFA deprotection?

Every peptide chemist will tell you that C-terminal amides are more stable than C-terminal acids. It turns out that is completely wrong for intracellular proteins! A fantastic collaboration with @jcornlab, @matthias_muhar, @jakobfarnung

We have a PhD position to work on protein SUMOylation using a mixture of approaches, funded by our ERC Starting Grant. Orléans is a beautiful medium-sized city on the Loire, not far from Paris. I'll do my best to make it a good experience.
