@FengYungWang1 @Aging_CEU @Nature That's a really good question, and one that we will hopefully be able to answer in the future. Our initial work on a limited number of Rapamycin treated mice shows that yes there might be a best time window for intervention. But GxDrug is the next step.

@DannyArends @Aging_CEU @Nature That's super big work and fantastic findings! One quick question i am curious is whether those "aging intervention" drugs (repaying, metformin) also have an optimal window for the intervention? may be beneficial in young but harmful in elders, or otherway around?

Candidate genes at other loci point to lysosomal function, autophagy, fatty acid oxidation as well as mTOR signalling. So immune function does feature, alongside broader metabolic and cellular maintenance pathways.

@zhaoweiasu @Nature Great question! The honest answer is we don't fully know why. The leading idea is that males and females face fundamentally different reproductive trade-offs that get wired into separate genetic networks over time. A steep price: for some loci yes, they help early and hurt late.

@DannyArends @Nature Congrats! Very interesting study! Any clue why epistatic links are non-overlapping between male and female? Does that mean we have paid a steep price for reproduction, but in totally different way for different sex?

@DannyArends @AllThingsApx Wow this is incredible! Very interesting! Note on the sex differences: Looking at exercise in rats, we were surprised that the *vast majority* of significant active chromatin and transcriptional changes were in only one sex. The common changes were only ~5%!

4/5 The result that surprised me most: The genetic interaction networks in males and females are almost completely separate 🤯. 78 epistatic links in males, 72 in females. Only 2 overlap. If you ignore sex in aging genetics, you get the wrong answer. Full stop.

@Aayush13138763 I really enjoy seeing code grow and adapt more features over time. The basic web server grew out of serving unsecure https 1.0 in a request response loop. Its current form does multiple HTTPS domains on 1 ip using SNI and ACME renewal, supports keep alive and streaming video.
