Mechanistically, redox state creates a self-reinforcing circuit linking HIF1α signaling to post-translational regulation of IDH1, a key metabolic enzyme. This circuit couples local oxygen status to ISC metabolic programming. 🔄🧪

We found that hypoxia + Wnt signaling collaborate to restrict NOX1—a ROS-generating enzyme—to the crypt base in the distal colon. This spatial control shapes where stem cells maintain an oxidative state needed for cell-cycle entry. 🔥
