The increased mutation rate of DNA-repair–deficient mutators accelerates resistance evolution in bacteria and cancer. In a great collaboration with @MulkernAdam, we find that DNA-damaging antibiotics can turn this advantage into a liability—but only for certain genotypes.

We paired antibiotics with DNA-damaging agents and evolved a panel of E. coli repair mutants using high-throughput evolution at EMBL. Some hypermutators were constrained, but the strategy only works if you match the damage to the specific repair defect...

This is an important step in unraveling the path of antisense antibiotics (asobiotics) across the complex envelope of gram- bacteria and on to finding their mRNA targets. I hope Adam and Marco have enough energy left to expand this to many more species, including gram+ 😉
