Fantastic work 👏 from @aaronmring lab showing specific binding of FDA-approved tx antibodies to unintended targets in 28% of Abs tested. E.g Pembrolizumab doesn’t bind PD-1 only but TDGF1 surface protein. This could lead to unexpected toxicity or mecanism of action.

@allisonoconn In a lot of trials looking at predictive value of target expression use primary tumors while metastatic lesions are treated. Discordance in target expression between primary tumors and marches mets is not uncommon, especially for targets with immune function

Today I learned that the current thinking for resistance to antibody-drug conjugates (ADCs) is that resistance is more likely to the payload than the target, so it might be possible a second ADC targeting the same thing but with a different payload would be effective!

@allisonoconn I think it is multifactorial, for instance drug uptake can be influenced by tumor/normal tissue target expression ratio, tumor vascularization and fibrotic microenvironment.
