In our latest work led by @firewhenreddy, we engineered ancestral glutamate transporters to track an evolutionary change in ion coupling. Using biochemistry+cryo-EM, we found that the key intermediate is the allosteric 'breaking' of ion coupling. biorxiv.org/content/10.110…
