New preprint on technologies to scale up CRISPR screens. We use them to map 665,856 pairwise genetic perturbations and outline a path to comprehensive interaction mapping in human cells. We also introduce an approach for cloning lentiviral libraries with billions of elements.

Really excited for the news today as Link Cell Therapies officially launches out of stealth with the announcement of their Series A. The company has been hard at work for several years turning a prototype Boolean-logic AND-gated CAR T cell into a real product with real potential to help patients. With LNK001 headed towards a Phase 1 trial in renal cell carcinoma in 2026, we are excited about the team Link has built - and we hope their work will redefine the universe of antigens and cancers we can target with CAR T cells. Today in @Nature we report a new prime editing strategy that can rescue a common cause of many genetic diseases in a disease-agnostic manner. This approach converts a redundant endogenous tRNA into an optimized suppressor tRNA, enabling a single prime edit to rescue premature stop codons across different diseases.
