Interested in conditional degron tags for your protein of interest? We are too! But after finding the discovery process slow and inefficient, we developed a new system to more robustly identify degron tags. 🧵 nature.com/articles/s4146…

Congrats @DanB0nD1 ! Great read for those interested in degron tagged systems. Happy to be a small part of it. Now let’s get more in vivo PK/PD for the compounds!

@JSheltzer Not what you were exactly going for but…for in vivo it’s hard to control the exposure concentrations to not inhibit both of X & Y.

Drug X has an IC50 value of 5nM against Kinase Y and an IC50 value of 50nM against Kinase Z. These values were measured both in vitro and in cellulo. Based on this data, can one reasonably claim that Drug X is highly selective for Kinase Y over Kinase Z?

Stoked that this work is out! It represents the awesome power of @CancerDepMap for understanding gene function and the promise for new cancer therapeutics. Congrats @DanB0nD1 and thanks to all our co-author friends and colleagues!

🎯 Exciting new target emerging from @CancerDepMap data XPR1, a phosphate exporter, is a dependency in ovarian and uterine cancers with high SLC34A2 expression Work led by @DanB0nD1 and @Biologenius in @DrFranVazquez and Todd Golub labs Full paper rdcu.be/cLE4k

Should data from large-scale RNAi screens be considered alongside CRISPR screens to identify cancer vulnerabilities? Our analysis suggests that RNAi screens can still provide useful and complementary information when CRISPR-Cas9 knockout is pan-lethal biorxiv.org/content/10.110…🧵⬇️

Amazing. This resource will be transformative for so many efforts, including those building off of my thesis work 🙌 @Biologenius @Sumaiyalqbal & @veronica_rendo, I'm counting on you to dig up some GEMINI targets in there! 💪😉

Pediatric cancers don’t have as many targeted therapies as adult cancers. “We felt like this effort to map genetic dependencies in pediatric cancers was all the more important because we need to identify new targets for these cancers.” @CancerDepMap broad.io/PediatricDepMap

Therapeutics targeting pediatric cancers unfortunately often lag behind adult cancers. Pleased to be a part of a group to address this disparity to highlight the many opportunities.
The Cancer Dependency Map@CancerDepMap

Pediatric cancer was underrepresented in @CancerDepMap. We teamed up with pediatric cancer investigators to profile cell lines from 13 pediatric tumors. Excited to share the results of the analysis led by @NeekeshD. 1/5 nature.com/articles/s4158…

Great across the pond collaboration to build a more comprehensive map of cancer dependencies! But we still have farther to go. If you have cell models and want to donate please reach out to @CancerDepMap #teamscience
The Cancer Dependency Map@CancerDepMap

In collaboration with @DepMapSanger, we develop computational methods to integrate Broad and Sanger CRISPR data to maximize their power. This resulted in the largest existing CRISPR currently with 869 cell lines! Read at doi.org/10.1038/s41467…. 1/2
