Excited to finally see this @DanaFarber_GU (@eddy_saad @MarcMachaalani) collaboration with @kaelin_lab @broadinstitute led by @RemailehAbu published in @NatureMedicine 1) PTHLH->HIF2 transcriptional target in ccRCC. 2) Increased PTHLH is necessary/sufficient for the induction of hypercalcemia & cachexia in preclinical models. 3) Two HIF2 inhibitors improved hypercalcemia and cachexia in patients with ccRCC. => Clinically, this can explain at least in part weight gain on HIF2 inh => Future, can PTHrP-targeted interventions (directly through anti-PTHrP therapies or indirectly through the HIF2–PTHrP axis) block the onset of cachexia?
