🧬 #ScienceSaturday ❓ Why do some colorectal cancers resist #immunotherapy, even when they should respond? ➡️ A new study in @CellCellPress explored how immune cells inside tumors interact and discovered that resistance to PD-1 blockade may be driven by suppressive macrophages and exhausted T cells. ➡️ Researchers found that successful responses were linked to coordinated activity between cancer-fighting T cells and immune-supporting macrophages that help activate anti-tumor immunity. ➡️ Tumors that resisted treatment showed higher levels of TREM2+ macrophages and multiple immune checkpoints, including LAG3 and CTLA4, creating an immunosuppressive tumor environment. ➡️ By combining PD-1 blockade with therapies targeting LAG3, CTLA4 and TREM2, researchers achieved complete tumor clearance in up to 100% of mismatch repair-deficient colorectal cancer models. 🌟 The findings highlight how reprogramming immune cell communication inside tumors could help overcome immunotherapy resistance and improve outcomes for more patients with colorectal cancer. 🔗 Read the study: cell.com/cell-reports-m… @BhardwajLab @CimenBozkus_C @matt_brown04 @NatalieVaninov @SpitzerLab @neoviral @rsamstein @IcahnMountSinai @SinaiImmunol @MountSinaiNYC

Viruses reprogram host cells by reshaping chromatin, but the epigenetic activities of most viral proteins remain unknown. In our new preprint, we use pooled single-cell epigenomic screening to profile 110 viral transcriptional regulators simultaneously, revealing widespread chromatin remodeling, unexpected functional convergence across diverse viruses, and shared targeting of NF-κB, AP-1, p53, and Sp1/KLF regulatory networks. So we blocked IFNγ in Mtb infected macaques and….the animals were fine. Turns out mice overestimate the importance of IFNγ in TB because IFNγ drives iNOS in mice but not macaques or humans. This means we don’t actually know how T cells suppress Mtb. rdcu.be/ff9GB
