@OWallerman @kirk3gaard @gaworj One trick you can try with low conc is to increase the conc by reducing the volume. It's a little tricky but you can do SRE with as little as 5-10 uL of sample.

@kirk3gaard @gaworj Try going in with as high a concentration as possible. The recovery efficiency for small fragments is heavily influenced by concentration.

@gaworj @circulomics I expected "losing" a lot of DNA but not all of it. Have used it successfully for keeping even shorter fragments in the past.

@OmicsOmicsBlog Thanks for this article. I wonder how much of this is due to incompatibility discovered only later. I extracted the best DNA I have ever extracted with Circulomics, and for some reason ULR sequencing failed, on several occasions, with PromethION. The disappointment was crushing.

The human genome contains some incredibly repetitive stretches of DNA that are hard to read (i.e., to sequence). @Genome_gov has been funding start-up companies to develop technologies for sequencing such complicated regions of DNA.
