❓ How can we turn T-cells into cancer-fighting “soldiers” without changing their DNA?
➡️ A new study in Nature Biotechnology introduces an all-RNA platform that can turn T-cell genes on or off using epigenetic CRISPR tools called CRISPRoff and CRISPRon. Instead of cutting or rewriting DNA, this method “reprograms” genes by adding or removing chemical marks — allowing researchers to boost or silence key immune functions safely.
➡️ The team showed this can enhance CAR T-cell therapies, helping immune cells better recognize and attack tumors while reducing potential side effects linked to traditional gene editing.

In a medical milestone, a customized base editor was developed, characterized in human and mouse cells, tested in mice, studied for safety in non-human primates, cleared by @US_FDA for clinical trial use, manufactured as a complex with an LNP, and dosed into a baby with a severe, rapidly progressing genetic disease... all in an astounding 7 months. Best of all, the infant patient shows apparent benefit. Congratulations to @kiranmusunuru, Rebecca Ahrens-Nicklas, and other team members for this heroic and inspiring effort, which has implications for the hundreds of millions of patients that suffer from thousands of genetic diseases.

➡️ Immunotherapies targeting neoantigens can be effective in mediating long-term durable responses in advanced cancer patients. However, identifying these neoantigens from metastatic tumors can be logistically challenging.
🧬 In this study, investigators looked at whether archived, preserved primary tumor samples could be used for neoantigen discovery.
➡️ The study found that primary FFPE tumor samples could be used to discover most of the neoantigens present in metastatic tumors and even unique neoantigens not present in metastatic tumors.
🌟 This approach would reduce the need for additional, sometimes complicated resections and expedite the time to treatment! These promising findings could pave the way for more accessible and personalized therapies.
