We have reported a series of first-in-class inhibitors for YEATS domain proteins since 2018, including an ENL inhibitor in 2018, an AF9 inhibitor in 2020, and now the first YEATS2 inhibitor. This work is now published in @nchembio nature.com/articles/s4158…

Congrats @Amgen @jaybradner and Dark Blue Therapeutics! Exciting to see ENL/MLLT1 biology advancing toward therapeutic translation in AML - a great example of how basic chromatin biology can inform new treatment strategies.

Thanks Bruce @LifeSciVC. Time cannot move fast enough to bring this brave idea and prototype medicine to patients with AML. As a stem cell transplant physician, I am well calibrated to the challenge AML patients face. All learnings, insights and leaders at @Amgen in ALL will be accessible to this amazing team of innovators.

Our latest paper has just been published in Cell! doi.org/10.1016/j.cell… We developed a new method called MCC ultra, which allows 3D chromatin structure to be visualised with a 1 base pair pixel size.

@Gandhar_Datar joined the lab when the world was shutting down, having just gotten started with guidance from @therealDrGundry and @BrunettiLab, who then moved on in their careers. Tough work in isolation until @RibackLab joined @bcmhouston, when informal chats got us thinking about the project in a whole new way. We considered how the aberrant function of NPM1c was related (or not) to its nucleolar WT function. A great collaboration grew, with Gandhar deeply learning both hematology and biophysics! Many ppl world-wide contributed and @sciencyelmira was the closer, getting the in vivo models working. Thanks to all, including the reviewers and editors for feedback.
