Our new Nature study, lead by @yuexing identifies a brief early-life window when skin dendritic cells sense common allergens and trigger inflammation right in the skin without migrating to lymph nodes. We define this in-situ activation state as peripheral immune inducer (pii)DC, and show it emerges because of a still-maturing HPA axis and low systemic glucocorticoids. #Naiklab 👉Key takeaways: 1. Early-life immunity plays by different rules. During this window, innate responses to allergens in skin are heighted. 2. Hormones shape immunity in real time. Developmental neuroendocrine states profoundly tune how the immune system reacts, and can set the stage for inflammatory disease. A great study by the Hoffmann lab in @JExpMed shows innate immune memory can be propagated in a cell-extrinsic way via local cytokine trapping. In our Insight, Luca Frosio and I discuss how these findings suggest spatial niche-specific contron of trained immunity 🎯
Journal of Experimental Medicine@JExpMed

Insights: Frosio and Ostuni @MyUniSR discuss study by Gorin et al. (hubs.la/Q043CjJY0) which reveals an unexpected mechanism by which IFN-γ sustains trained immune states through prolonged signaling driven by cytokine retention at the cell surface. hubs.la/Q043CjzR0

Today, we see the fruit of 5+ years of teamwork. We have built a map of the neutrophil compartment (mostly in mice - but also humans). It provides a biological structure that focuses on a single immune cell lineage to discover the many ways in which neutrophils to physiology.

I’m thrilled to introduce you to the NeuMap! our latest work in @Nature. A global, comprehensive, single-cell transcriptional atlas of neutrophils across 47 biological conditions in human and mice. A real tour-de-force 🗺️ @AndrsHidalgo16 nature.com/articles/s4158…

Finally out! In our last work, we show that diet-derived AhR ligands 🥦 are required for the optimal efficacy of anti-PD1 therapy promoting reinvigoration of progenitor exhausted CD8 T cells and licensing the functional response of effector CD8 T cells. nature.com/articles/s4146…

Glad that our paper is officially out @CellCellPress. BRD4 condensate functions as a pH sensor to tune inflammatory response in macrophage. cell.com/cell/fulltext/… earlier post: x.com/XuZhouLab/stat… and could-have-been cover by my student&artist Cathleen Krabak @harvardmed

Beyond excited to share our #NewPaper in @CellCellPress! Inflamed environments acidify intracellular pH. BRD4 senses this via transcriptional condensates, tuning #macrophage responses to match demand and consequences of inflammation. #inflammation #pH authors.elsevier.com/sd/article/S00…

My biggest adventure (yet): the Tor Des Geants, one of the hardest ultra-trail races❤️‍🔥. Can't express the mix of happiness, fear & fatigue of running 113 hours (350km, 25K climbing) in the mountains. Even made it in the top 15%! But now.. back to work, cool science is cooking 👨‍🔬

Out in @ScienceTM! The Naldini lab delivers another masterclass of stem/immune cell gene engineering to deliver cytokines in the TME and boost CAR-T cell activity in glioblastoma. Kudos to @f_rossari @giorgiaalvisi and Nadia Coltella for this important study🔥
Federico Rossari@f_rossari

🧵1/ 🚨 Just out in Science Translational Medicine @ScienceTM our latest work @NaldiniLab: We show how tumor-targeted cytokines can rescue CAR-T function and recruit endogenous T cells to fight glioblastoma, the toughest brain cancer. 🔗science.org/doi/10.1126/sc… 👇 Here's a thread:
